Barcelona-El Prat is close enough to the city that the airport transfer should be simple, but the best route depends on your terminal, hotel area, luggage, and arrival time. The mistake is choosing a route before checking where your first night actually is.

For most first-time visitors, Aerobus is the easiest public option if you are staying near Placa Catalunya, Placa Espanya, or a simple metro connection. The train is useful from Terminal 2 if your hotel is near Sants or Passeig de Gracia. The metro works better for some outer connections than for a direct old-town arrival.
If you land late, travel with children, or stay on a narrow old-town street, a taxi or pre-arranged transfer can be worth it.

Aerobus links the airport terminals with central stops such as Placa Espanya, Gran Via/Urgell, Universitat, and Placa Catalunya. It is easy to understand after a long flight and has luggage space, which is why it works well for many first arrivals.
The airport train uses the Rodalies R2 Nord route from Terminal 2. It is practical for Sants and Passeig de Gracia, especially if your hotel is in Eixample. If you land at Terminal 1, include the terminal connection in your timing.
The L9 Sud metro serves both terminals but does not go directly into the historic center. It can be useful if your accommodation connects well through Torrassa, Collblanc, or Zona Universitaria.
The regular city bus and night bus can be useful for budget travelers, but they are less beginner-friendly with luggage.

If you are staying in Eixample, check whether Passeig de Gracia, Urgell, Universitat, or a nearby metro station makes the final walk simple. For El Born or the Gothic Quarter, avoid assuming the closest stop is easy with bags. Some streets are narrow, busy, or awkward late at night.
